Background

The DAB kit gives a brown reaction product with peroxidase. Sections that have been stained with DAB can be covered in organic covering medium (e.g. HistoPerm LINARIS Cat. No. E6001).

The reagents are sufficient for approx. 1000 tissue sections.

  • 1 bottle reagent 1 (Substrate Buffer) 100 ml
  • 1 bottle reagent 2 (DAB) 6ml

Preparation of the working solution

  • Add 4 drops of DAB Chromogen Solution (DAB Concentrate) to 5 ml of DAB Substrate Buffer and mix well.

Use solution for up to 6 hours!

  • Rinse tissue with wash buffer after the previous incubation step.
  • Apply the desired amount of working solution to the prepared tissue section.
  • Incubate for 5 – 15 min at room temperature.
  • Rinse with dist. or deionized H2O.
  • Counterstain with hematoxylin (depending on strength) for 30 sec to 5 min.
  • Rinse with dist. or deionized H2O.
  • Blueing in tap water for at least 5 minutes.
  • Dehydrate in the ascending alcohol series and from the xylene permanently cover.

Note: DAB can also be covered with water. The staining result is brown stained tissue areas where the peroxidase is located.
