Background

Steroid hormone receptors are of great importance for the diagnosis and therapy of breast cancer. The expression of estrogen or progesterone receptors in tumor tissue is a prerequisite for a patient’s response to anti-hormone therapy.

The monoclonal antibody E051 was obtained from culture supernatant of mouse hybridoma cells and is available in ready-to-use form.

The monoclonal antibody E051 reacts with the N-terminal sequence of the human estrogen receptor (ER). It shows nuclear staining of epithelia of the uterus and mammary glands.
Specifically, estrogen receptors are experimented in the nuclei of the corresponding carcinomas. Tissue-dependent cytoplasmic staining may also occur.
The antibody can be used for frozen sections as well as for formalin-fixed kerosene material. A prerequisite for satisfactory staining results is a first-class preservation status of the tissue. However, recent investigations by our development laboratory have shown that differences in fixation can be largely compensated for by pretreating the tissue sections with our HistoSafeENHANCER (LINARIS CatNo E7000). After this pretreatment, a one-hour incubation period with the primary antibody (just as with frozen sections) is sufficient.

Pretreatment

Pretreat paraffin sections for 20 min at 96-100°C with HISTOSAFE ENHANCER (LINARIS CatNo E7000) in a water bath or 3 times 6 min in the microwave. Cryostat material does not require pretreatment!

Incubation Period

1 hour by room temperature

Usage

Ready-to-use in PBS, BSA, NaN3 (0,09%) pH 7,4(*) suitable on cryostat sections and on formalin-fixed, paraffin-embedded tissue sections.
