Background

AEC is a commonly used chromogene for immunohistochemical procedures. Upon reaction with peroxidase AEC produces a red reaction product that is soluble in alcohol. Thus, aqueous counterstain (e.g. Hematoxylin LINARIS CatNo. EGH3411) and mounting medium (e.g. LinMount LINARIS CatNo E6004) must be used.

  • 1 drop bottle reagent 1 (Buffer solution) 2 ml
  • 1 drop bottle reagent 2 (AEC) 2 ml1 drop bottle reagent 3 (H2O2) 2ml
  • 1 drop bottle reagent 3 (H2O2) 2ml
  • 1 drop bottle (empty) for the working dilution (G)

Preparing the working dilution (sufficient for approximately 100 sections)

  • 10 ml Aqua dest. in bottle G
  • Adding of 2 drops Reagent 1, mix well
  • Adding of 2 drops Reagent 2, mix well
  • Adding of 2 drops Reagent 2, mix well

Working solution used directly!

  • Incubate tissue sections with the substrate until suitable staining develops
  • 20 – 30 min incubation at room temperature and protected from light provides a good staining intensity.
  • Wash with Aqua dest.
  • Mount slides with aqueous mounting medium (e.g. LinMount LINARIS CatNo. E6004).
