Background Human CD45RO

The monoclonal antibody E027 (UCHL1) recognizes a T cell-associated antigen with a molecular weight of approximately 180 kDA. This is the low molecular weight isoform of the leucocyte common antigen (LCA) CD45, which since the IV International Workshop on Leucocyte Differentiation Antigens (Vienna 1989) has been designated CD45RO). The antigen is expressed by a functional subset of CD4+ T cells, by most thymocytes, and by granulocytes and monocytes. Studies of B- and T-cell lymphomas and plasmocytes suggest that CD45RO (UCHL1) is expressed in normal B cells prior to differentiation to plasma cells, but plasmocytomas are negative except for a few reactive lymhpozytes. Negative for CD45RO are also cutaneous T-cell lymphomas (mostly CD4+, CD45R-).

Synonyms

T cell Marker, CD45RO, UCHL1

Reagents sufficient for about 50-100 tissue sections

  • 1 dropper bottle HISTOPRIME® T cells (CD45RO, UCHL1) (Bottle, 5 ml)

Antigen

IL-2-activated T cells (CA1)

Specificity

Thymocytes, CD4+/CD45RO+ T-lymphocytes (T-helper cells)

Abnormal Tissue

Differentiation of T-cell lymphomas. UCHL1 is negative in plasmocytomas and most T-cell lymphomas in the skin; Reed-Sternberg cells are also negative in Hodgkin lymphoma.

Clone

UCHL1

Isotype

Mouse lgG2a

Fusion Partners

E027 is produced from the cell culture supernatant of mouse hybridoma cells secreting UCHL1. An IL-2-dependent T cell line (CA1) was used for immunization.

Characterization of B-cell and T-cell lymphomas in cryostat material and paraffin-embedded tissue.

Pretreatment

Pretreatment of formalin-fixed material with Pronase is not required.

Incubation Period

1 hour at room temperature

Usage

Ready-to-use in PBS, BSA, NaN3 (0,09%) pH 7,4(*) suitable on cryostat sections and on formalin-fixed, paraffin-embedded tissue sections.

Control Tissue

Tonsils, lymph nodes, spleen

Recommended Secondary Reagents

Alkaline Phosphatase Vectastain® ABC Mouse IgG (Vector CatNo AK-5002) and SubstrateKit e.g. Vector® Red (Vector CatNo SK-5100).
Peroxidase Vectastain® ABC-Elite Mouse IgG (Vector CatNo PK-6102) and Peroxidase Substrate-Kit e.g. DAB (LINARIS CatNo E108) or HistoGreen (LINARIS CatNo E109 ).
