Background Macrophages/Monocytes

The monoclonal antibody recognizes the cytoplasmic antigen L1 (calprotectin) of the monocyte/macrophage lineage. It is expressed by neutrophil granulocytes as well as tissue histiocytes, alveolar macrophages, Kupffer cells, splenic macrophages, infiltrating macrophages, sinoidal macrophages as well as epithelial giant cells. It is also present in the cytoplasm of epthelial cells.

Specificity

Cytoplasmic antigen in cells of the monocyte/macrophage lineage (including histiocytes, alveolar macrophages, Kupffer cells, splenic macrophages, tumor-infiltrating macrophages, sinoid macrophages, and epithelial giant cells; also granulocytes under certain conditions.

Fusion Partners

BALB/c mice were immunized with purified blood monocytes. Spleen cells from these animals were fused with mouse myeloma cell line NS-1. The monoclonal antibody is available in readyto-use form.

The monoclonal antibody E026 (MAC 387) stains the cytoplasm of cells of the monocytemacrophage lineage. For example, infiltrating and reactive histiocytes or tumor-infiltrating macrophages can be detected.

Pretreatment

proteolytic pretreatment with 0.1% Pronase (LINARIS CatNo E110) 10 min at room temperature

Incubation Period

1 hour by room temperature

Usage

Ready-to-use in PBS, BSA, NaN3 (0,09%) pH 7,4(*) suitable on cryostat sections and on formalin-fixed, paraffin-embedded tissue sections.
