Background

Steroid hormone receptors are of great therapeutic importance in the treatment of breast cancer. The expression of estrogen or progesterone in tumor tissue is a prerequisite for a patient’s response to anti-hormone therapy.

Reagents sufficient for about 50-100 tissue sections

The monoclonal antibody E054 was obtained from culture supernatant of mouse hybridoma cells and is available in ready-to-use form.

The monoclonal antibody E054 reacts with the human progesterone receptor (PRG). It shows a nuclear staining of cell nuclei of the corresponding carcinomas expressed. Depending on the tissue, cytoplasmic staining may also occur.
The antibody can be used for frozen sections as well as for formalin-fixed kerosene material. A prerequisite for satisfactory staining results is a first-class preservation condition of the tissue.
In a series of tests in our development laboratory, pretreatment of the deparaffinized sections with the HistoSafe enhancer (CatNo E7000) has proven to be the best prerequisite for good staining results. After this pretreatment, a one-hour incubation time with the primary antibody (the same as for frozen sections) is sufficient.
With an incubation time of 16 h at room temperature without pretreatment, useful results can also be obtained.

In addition, we offer other paraffin-embedded antibodies such as anti-estrogen receptor (LINARIS Cat. No. E051), anti-cathepsin D (LINARIS Cat. No. E048) or anti-p53 (LINARIS Cat. No. E052) for questions on breast carcinoma.

Pretreatment

Pretreat paraffin sections for 20 min at 96-100°C with HISTOSAFE ENHANCER (LINARIS CatNo E7000) in a water bath or 3 times 6 min in the microwave. Cryostat material does not require pretreatment!

Incubation Period

1 hour at room temperature

Usage

Ready-to-use in PBS, BSA, NaN3 (0,09%) pH 7,4(*) suitable on cryostat sections and on formalin-fixed, paraffin-embedded tissue sections.
