Background

Prostate specific antigen is a glycoprotein with a molecular weight of 33,000-34,000 daltons. PSA is present exclusively in prostate gland tissue and seminal fluid and is thus distinct from prostate specific acid phosphatase (PAP). It is detectable in normal, benign, hyperplastic as well as in cancerous prostate gland epithelium.

Reagents sufficient for about 50-100 tissue sections

  • 1 dropper bottle HISTOPRIME® PSA (Bottle, 5 ml)

Antigen

Human prostate specific antigen

Specificity

Secretory and ductal epithelium in normal and neoplastic prostate tissue.

Clone

ER-PR8

Isotype

Mouse lgG1

Fusion Partners

The monoclonal antibody was obtained from culture supernatant of mouse hybrids.

The monoclonal antibody against PSA reacts with secretory and ductal epithelium of the prostate in normal and neoplastic tissue. In a clinical study, 95% of prostate carcinomas were detected as positive with the antibody clone (usually the antibody does not react with bladder, stomach, colon, kidney, pancreas and lung or carcinomas of the corresponding tissues.

Pretreatment

Pretreatment with proteolytic enzymes is not required.

Incubation Period

1 hour by room temperature

Usage

Ready-to-use in PBS, BSA, NaN3 (0,09%) pH 7,4(*) suitable on cryostat sections and on formalin-fixed, paraffin-embedded tissue sections.

Control Tissue

Prostate

Recommended Secondary Reagents

Alkaline Phosphatase Vectastain® ABC Mouse IgG (Vector CatNo AK-5002) and SubstrateKit e.g. Vector® Red (Vector CatNo SK-5100).
Peroxidase Vectastain® ABC-Elite Mouse IgG (Vector CatNo PK-6102) and Peroxidase Substrate-Kit e.g. DAB (LINARIS CatNo E108) or HistoGreen (LINARIS CatNo E109 ).
