Background

Vimentin is a protein with a molecular mass of 57kD. It forms, together with microfilaments and microtubules, the cytoskeleton of non-epithelial cells, e.g. non-muscular mesenchymal cells and cells of mesenchymal origin including vascular smooth muscle, astrocytes and numerous cultured cell lines. It frequently occurs together with desmin.
Expression of this protein is retained during the conversion of normal to neoplastic cells.

BALB/c mice were immunized with vimentin (from pig lentils). Spleen cells from these animals were fused with a tumor cell line. The resulting hybridoma cells were used for ascites production.

The monoclonal antibody E034 reacts with human vimentin as well as with vimentin in pig, chicken and rat. It recognizes tumors of mesenchymal origin, e.g. sarcomas and malignant lymphomas.
Importantly for its application, this antibody does not require pretreatment with pronase. For formaldehyde-fixed material, the intensity of the staining may be indicative of the fixation time.

Pre-treatment with proteolytic enzymes not required!
HISTOAFE-ENHANCER (LINARIS CatNo. E7000 can improve the results).

Incubation Period

1 hour at room temperature

Usage

Ready-to-use in PBS, BSA, NaN3 (0,09%) pH 7,4(*) suitable on cryostat sections and on
formalin-fixed, paraffin-embedded tissue sections.
