Background

The factor VIII complex is a protein found in normal human plasma and belongs to the coagulation factors. It consists of 3 subunits: factor VIII RCF (ristocitin cofactor), factor VIIIC (coagulant portion) and factor VIII-associated antigen (Von Willebrand factor). This represents the large-molecule portion of the factor VIII complex. Its formation occurs in blood platelets and in the endothelium of all vascular segments, probably also in the RES of the liver. It is also found in megakaryocytes.

Fusion Partners

(C57BL/6xBalb/c) F1 mice were immunized with factor VIII-associated antigen. Splenocytes from these animals were fused with myeloma cell line NS1. The antibody is available as culture supernatant of the resulting hybridoma cells.

The monoclonal antibody against Von Willebrand factor E033 is used to stain (cytoplasmic, granular) endothelial cells and megakaryocytes in human tissue. Tumors originating from endothelial cells or megakaryocytes can be deployed by detecting this antigen. In addition, vascularization within different tissues can be assessed and tumor invasion into blood vessels can also be detected.

Pretreatment

Proteolytic pretreatment with 0.1% Pronase (Cat.No.E110), 10 minutes at room temperature.

Incubation Period

1 hour at room temperature

Usage

Ready-to-use in PBS, BSA, NaN3 (0,09%) pH 7,4(*) suitable on cryostat sections and on
formalin-fixed, paraffin-embedded tissue sections.
